Antibody Framework Reengineering Technology

Our proprietary framework-reengineering technology developed by our scientific founder compares favorably to other humanization methodology in reducing the immunogenicity of a monoclonal antibody without affecting its target specificity and affinity. Humanized antibodies developed based on this method belong to a new class of antibodies, termed Framework Reengineered antibodies with over 95% human origin. We believe that the uniqueness and novelty of our technology offer SinoMab with global competitive edge that rivals other US and European biotechnology companies in developing safer, and more efficacious therapeutic antibodies.

Our proprietary Framework Reengineered antibodies are designed based on structural sequence information from promising murine antibody candidates. Each individual framework sequence of the murine antibody is dealt with separately and independently, and replaced by the corresponding framework sequence of a human antibody, without modifying the CDR sequences, using a set of criteria. The framework reengineering technology has certain advantages over existing humanization technologies such as CDR grafting in several areas:

  • Flexible method that lead to speed and higher success rate
    The framework reengineering technology provides flexibility in the choice of framework sequences, increasing the chances of success in maintaining the original specificity and affinity after the re-engineering;

  • Substantially reduce potential immunogenicity
    The framework reengineering technology minimizes the need to introduce framework amino acids derived from the parent antibody, thereby reducing the chances of creating new T- and B- cell epitopes in the resultant FR-Patched antibodies that will be substantially non-immunogenic when used in human, with substantial retention of the specificity and affinity of the parent immunoglobulin to the antibody.
